English Abstract In May 1994 a collaborative study for food microbiology
was organized to test the newly developed reference material containing
Clostridium perfringens (strain D10) spores. The objective of this
international collaborative study was to evaluate the suitability of the
reference material for quality control of the enumeration of Clostridium
perfringens as described in standard methods for food microbiology used in
EU countries. Twenty-six European laboratories participated in the
collaborative study. In each laboratory 2 series each of 6 capsules were
tested on two different days by the same technician. Each series of
capsules contained one uncontaminated capsule. The capsules were
reconstituted according to a standardized procedure and analysed in
duplicate using three culturing methods. Two methods were standardized for
all laboratories one based on the ISO 7937 (Anonymous, 1985) using a
selective Tryptose-Sulphite-Cycloserine agar (TSC-SM) and one using a non
selective Sheep Blood agar (SBA-SM). The third method was chosen by the
participating laboratory (OWN). There were few technica problems in the
participating laboratories. Several laboratories found a pH value above the
pH requirements for the TSC-SM medium and/or SBA-SM medium. However these
deviations in pH appeared to have no effect on the counts of Clostridium
perfringens.The counts obtained with the SBA-SM were on average the
highest, which is probably due to the better recovery of Clostridium
perfringens on a non selective medium. Compared to BCR/food trials 5 and 6
the range of counts found by the different laboratories was quite small for
all three methods, indicating that the variation between laboratories was
small. This was the first trial in which the same material was examined in
two series on different days. A series effect was demonstrated for all
three methods. For the two standardized methods(TSC-SM and SBA-SM) a
learning effect was demonstrated between the first and second series of
analysis: both the T2/(I-1) values and the geometric means were closer to
each other for the second series than for the first series. This learning
was not observed for the OWN method. A reproducibility value within a
laboratory (Rl), in which the series component is included, was introduced,
to obtain an indication of the real within laboratory precission. The
values found for the repeatability (r), reproducibility within a laboratory
(Rl) and reproducibility between laboratories (R) were close to the
theoretical optimum, thus confirming good performance by the laboratories in
BCR/Food trial 7 as well as good quality of the reference material. The
Clostridium perfringens reference material proved to be stable during 15
months storage at -20 degrees C when enumeration was performed on the
selective TSC medium, whereas a decrease in contamination level was noted
with the non selective SBA medium. It is not clear what caused this
difference. During 4 weeks storage at elevated temperatures an acceptable
level of stability was achieved at 22 degrees C on both the TSC and SBA
medium. The reference material was less stable at 30 degrees C and 37
degrees C after 4 weeks storage, but due to the design of the challenge test
no conclusions could be drawn about the stability of the materials at these
temperatures during a transport of maximum 7 days. However the transport
times in the collaborative study, ranging from 1 to 7 days, appeared to have
no effect on the count results. The use of pour plates for the examination
of Clostridium perfringens reference materials might give problems with
counting due the merging of the black halos surrounding the colonies,
therefore in those cases the use of spread plates is recommended. It is
concluded that the Clostridium perfringens reference material gave
reproducible results and is therefore suitable as a reference material.
This reproducibility combined with the stability during storage at -20
degrees C of the reference materials, confirmed that the material was
suitable for a certification study.

In mei 1994 werd een ringonderzoek voor
levensmiddelenmicrobiologische laboratoria georganiseerd om een nieuw
ontwikkeld referentiemateriaal met Clostridium perfringens te testen. De
belangrijkste doelstelling van dit internationaal ringonderzoek was de
evaluatie van de geschiktheid van het Clostridium perfringens
referentiemateriaal voor het tellen van Clostridium perfringens volgens
standaardmethoden die in de EU landen gebruikt worden. Aan het
ringonderzoek namen 26 Europese landen deel. Bij elk laboratorium heeft een
analist 2 series van elk 6 capsules op 2 verschillende dagen getest. In
beide series was een capsule blanco. Elke capsule werd opgelost volgens een
voorgeschreven methode en in duplo geanalyseerd, waarbij drie kweekmethoden
gebruikt werden. Twee methoden werden standaard door alle laboratoria
uitgevoerd, een gebaseerd op de ISO methode 7937 gebruikmakend van selectief
Tryptose-Sulphite-Cycloserine agar (TSC-SM), in de andere standaardmethode
werd het niet-selectief Sheep Blood agar gebruikt (SBA-SM). De derde
methode werd door het deelnemende laboratorium zelf uitgekozen (OWN). Er
werden slechts weinig technische problemen waargenomen bij de deelnemende
laboratoria. Verschillende laboratoria vonden een Ph waarde voor het TSC-SM
en/of SBA-SM medium dat niet voldeed aan de gestelde Ph-eis. Deze Ph
afwijkingen lijken echter geen effect te hebben op de tellingen.De
tellingen met de SBA-SM lagen gemiddeld genomen het hoogst, wat
waarschijnlijk veroorzaakt wordt door de betere 'recovery' van Clostridium
perfringens op een niet selectief medium. De deelnemende laboratoria vonden
voor alle drie de methoden onderling overeenkomende tellingen. In dit
ringonderzoek werden de capsules voor de eerste keer in 2 series op 2
verschillende dagen getest. Voor alle drie de methoden werd een serie
effect aangetoond. Voor de twee standaardmethoden (TSC-SM en SBA-SM) bleek
een leereffect op te treden tussen de eerste en tweede analyseserie. Dit
leereffect kwam tot uiting in het dichter bij elkaar liggen van zowel de
T2/(I-1) waarden als de geometrische gemiddelden in de tweede serie. Er
werd geen leereffect waargenomen voor de OWN methode. Een waarde voor de
reproduceerbaarheid binnen een laboratorium (Rl), waarin de serie component
is opgenomen, werd geintroduceerd om een indruk te krijgen van de werkelijke
nauwkeurigheid in een laboratorium. De gevonden waarden voor de
herhaalbaarheid (r), reproduceerbaarheid binnen een laboratorium (Rl), en
reproduceerbaarheid (R) benaderden de theoretisch optimale waarde. Dit
toont aan dat de laboratoria in dit ringonderzoek goed presteerden en dat
het referentiemateriaal van goede kwaliteit is. Het Clostridium perfringens
referentiemateriaal is op het selectieve TSC medium stabiel gedurende 15
maanden opslag bij -20 graden C, terwijl gedurende deze periode een afname
in het besmettingsniveau werd waargenomen op het niet selectieve SBA medium.
Het is niet duidelijk wat de reden van dit verschil is. Bij 4 weken opslag
bij hogere temperaturen was alleen de stabiliteit bij 22 graden C acceptabel
op beide media. Het referentiemateriaal was minder stabiel bij 30 graden C
en 37 graden C, maar gezien de opzet van de test konden er geen conclusies
getrokken worden over de stabiliteit van het materiaal tijdens transport van
maximaal 7 dagen. In het ringonderzoek bleek echter dat de transporttijd,
die varieerde van 1 tot 7 dagen, geen invloed had op de tellingen.
Aangezien het Clostridium perfringens referentiemateriaal telproblemen kan
geven bij gebruik van gietplaten door spreiding van de zwarte zones rondom
de kolonies, wordt in die gevallen geadviseerd spatelplaten te gebruiken.
Geconcludeerd kon worden dat het Clostridium perfringens referentiemateriaal
reproduceerbare resultaten geeft en daarom geschikt is als
referentiemateriaal. Mede gezien de stabiliteit tijdens opslag bij -20
graden C van het Clostridium perfringens referentiemateriaal is dit
materiaal geschikt voor certificering.
