English Abstract The testing of a method to increase the sensitivity of
the current culture method used until now has been described. A sensitivity
increase will lead to a better estimation of the virus content of surface
water and to a better insight in measurements that should be taken to
minimise the infection risk represented by drinking water or bathing water.
In six out of 12 naturally contaminated water concentrates the virustiters
found by the immunoperoxidase method appeared to be higher, in five out of
12 the titers were comparable and in one out of 12 the titer was lower than
that found when the plaque overlay was used. The overall difference was not
statistically significant. So the desired increase in sensitivity of the
immunoperoxidase assay compared to the plaque overlay assay could not be
realised. Due to the minimal increase in sensitivity combined with the
smaller sample volume that can be processed, the immunoperoxidase method can
only be used when analysing water concentrates with a virus content of at
least 1 virus per milliliter.
Rapport in het kort
In dit rapport wordt een mogelijkheid beschreven om de
gevoeligheid van de huidige virus detectiemethode, de plaque overlay, te
verbeteren, wat zou leiden tot een betere inschatting van het virusgehalte
in oppervlaktewater. De immunoperoxidase methode bleek, voor het
kwantitatief aantonen van virussen, in de helft (6/12) van de onderzochte
waterconcentraten gevoeliger te zijn dan de plaque overlay methode. In vijf
van de 12 monsters waren de titers gelijk terwijl in een monster een hogere
titer werd gevonden met de plaque overlay methode. De verschillen tussen de
immunoperoxidase test en de plaque overlay test bleken statistisch echter
niet significant te zijn. Bijgevolg was het niet mogelijk om de gewenste
toename in detectiegevoeligheid van de immunoperoxidase test t.o.v. de
plaque overlay test te realiseren. Daar in de praktijk slechts een beperkt
volume van een totaal waterconcentraat met de immunoperoxidase methode
onderzocht kan worden, betekent dit dat de methode alleen geschikt is voor
concentraten met een hoog virusgehalte zoals oppervlaktewater en in- of
effluent van rioolwaterzuiverings installaties.
