Suppression of Plesiomonas shigelloides growth in the Direct Plating method for the enumeration of Escherichia coli in water

English Abstract
Plesiomonas shigelloides appeared to produce false positive reactions in the Escherichia coli Direct Plating (DP) method. In this study the DP method was modified by addition of the vibriostatic agent O/129 (2,4-diamino-6,7,diisopropylpteridinephosphate) in order to suppress Plesiomonas growth.In laboratory experiments, addition of 25 or 50 mg/l O/129 to Trypton Bile Agar (TBA) had no effect on either colony morphology nor counts of the test strain E. coli WR1. Pure cultures of the tested Plesiomonas strains (n=8) formed smaller colonies on TBA with O/129 than on TBA without O/129. These cultures formed indole negative pinpoints on membrane filters incubated on TBA with O/129. Plesiomonas colony counts declined: on TBA with 25 mg/l O/129 (TBA25) average log reduction was 3.1 , on TBA with 50 mg/l O/129 (TBA50) it was 3.8.Field studies (performed by three laboratories), in which 28 natural samples were analysed, showed that 50 mg/l O/129 reduces background growth and inhibits the growth of the indole positive, oxidase positive organisms (presumptive Plesiomonas). The number of indole positive, oxidase negative organisms (presumptive E. coli) increased.From TBA and TBA50, colonies were isolated and typed by API (Rapid 20E and 20E) identification systems. Of the indole positive, oxidase negative strains isolated in the Netherlands from both media 89-99% could be confirmed as E. coli. The confirmation rate of Hungarian isolates was lower, but increased from 61 to 74% when O/129 was used. Ninety-two percent of the indole positive, oxidase positive strains from TBA isolated in the Netherlands and 46% of these strains isolated in Hungary could be confirmed as Plesiomonas. In the Netherlands indole positive, oxidase positive organisms were not present on TBA50.

In het hier beschreven onderzoek is geprobeerd de groei van Plesiomonas te onderdrukken door toevoeging van het vibriostaticum O/129 (2,4-diamino-6,7,diisopropylpteridinefosfaat). Aantallen Plesiomonas kolonies namen af: op TBA met 25 mg/l O/129 (TBA25) was de gemiddelde log reductie 3.1, op TBA met 50 mg/l O/129 (TBA50) was deze 3.8.Veldonderzoek (uitgevoerd door drie laboratoria), waarbij 28 natuurlijke monsters werden onderzocht, toonde aan dat 50 mg/l O/129 de achtergrondgroei reduceert en de groei van indol positieve, oxidase positieve organismen (verdacht Plesiomonas) remt. Het aantal indol positieve, oxidase negatieve organismen (verdacht E. coli) neemt toe.Van TBA en TBA50 werden kolonies geisoleerd en getypeerd met API (Rapid 20E en 20E) identificatie strips. Van de indol positieve, oxidase negatieve stammen die in Nederland van beide media werden geisoleerd kon 89-99% bevestigd worden als E. coli. Het bevestigingspercentage van de Hongaarse stammen was lager, maar nam toe van 61 naar 74% bij gebruik van O/129. Twwe-en-negentig procent van de Nederlandse en 46% van de Hongaarse indol positieve, oxidase positieve isolaten van TBA werd bevestigd als Plesiomonas. Op TBA50 werden in Nederland geen indol positieve, oxidase positieve organismen meer aangetroffen.
