English Abstract One of the tests performed at the Department of
Biologicals of the Laboratory for Medicins and Medical Devices is an assay
for the detection of residual animal serum albumin (bovine serum albumin and
ovalbumin) in vaccines. The method used is an immunoprecipitation test
using Counter Current Electroforesis (C.C.E.). After electroforesis the
precipitate of albumin and anti-albumin serum is stained by Coomassie
Brilliant Blue. Traditionally the agarose-gels are visually inspected for
the presence of stained precipitates using the light of a bulb shining
through opal glass. This report describes the validation of a new detection
technique. The agarose-gel is illuminated using two halogen lamps. Under
this illumination the precipitates appear as little red lines.Using the
traditional detection-method (light bulb) the detection-limit was 1 mul
animal serum per litre vaccine. Using the new detection-method
(halogen-lamps) the detection-limit is lowered by at least a factor 2.
Furthermore, as a consequence of the elimination of background staining
artefacts the specificity of the detection is improved.

Een van de controle-testen op bepaalde vaccins is de
bepaling van residueel albuminegehalte m.b.v. Counter Current Electroforese
(C.C.E.).C.C.E. is een techniek die gebaseerd is op electroforese,
gecombineerd met immuno-precipitatie. Bij behandeling van de, in de test
gebruikte, agarose-gelplaten met coomassie briljant blauwoplossing, worden
de gevormde precipitaten donkerblauw gekleurd. Voor het aflezen van de
agarose-gelplaat wordt, in de standaard testmethode, gebruik gemaakt van een
detectiekast voorzien van een melkglasplaat waarin een verlichtingsbron
(gloeilamp) is aangebracht. Gebleken is dat de precipitaten op een
agarose-gelplaat van kleur veranderen, indien zij onder een bepaalde hoek
worden beschenen met behulp van halogeenlicht. Hierbij wordt de
detectie-grens met tenminste een factor 2 verlaagd. De detectie-grens van
de referentie lag op basis van de oude afleesmethode op 1 mul albumine per
liter runderserum. Daarnaast worden artefacten ten gevolge van aspecifieke
achtergrondkleuring vermeden, waardoor de specificiteit van de detectie
verbeterd is.
