English Abstract
In this report an overview is given of structure-activity relationships (SARs) described in literature that can be helpful for the daily human hazard evaluation of chemicals. SARs describe the relation between molecular structure and biological- or physical-chemical activity of the chemical. Chemicals that share structural features are presented that can have an effect on the toxicological endpoints: irritation, sensitisation, neurotoxicity (acetylcholinesterase-inhibition), genotoxicity and reproductive and developmental. These SARs were discussed in a Workshop. The results of this Workshop, which was attended by RIVM and TNO risk assessors and experts, are presented also. One of the results is a list of structural features with possible effects on the above mentioned toxicological endpoints. It is proposed to test these SARs in the next two years on chemicals for which experimental data are available. This report may help to increase the quality of the hazard assessment.

Rapport in het kort
In het rapport wordt een overzicht gegeven van de in de literatuur beschreven humaan toxicologische structuuractiviteitsrelaties (SAR's) en de bruikbaarheid van deze SAR's voor de dagelijkse humaan toxicologische evaluatie van stoffen. SAR's beschrijven de relatie tussen moleculaire structuur en de biologisch-chemische of fysisch-chemische activiteit. Voor de toxicologische eindpunten irritatie, sensibilisatie, neurotoxiciteit (acetylcholinesterase remming), genotoxiciteit en reprotoxiciteit zijn SAR's beschreven. Deze SAR's zijn besproken in een Workshop. De resultaten van de Workshop, waarbij deskundigen van zowel het RIVM als TNO aanwezig waren, zijn in dit rapport meegenomen. E5n van de resultaten is een lijst met structuurfragmenten met mogelijke effecten op de hierboven beschreven toxicologische eindpunten. Het wordt aanbevolen om deze SAR's de komende twee jaren uit te testen op stoffen waarvoor dit soort informatie reeds beschikbaar is en het mogelijke gebruik van de SAR's in de toekomst te evalueren. Dit rapport kan helpen om de kwaliteit van de effectbeoordeling te verbeteren.
