English Abstract A collaborative study has been carried out to establish
the precision and accuracy of 5 test systems for the assessment of the
toxicity of whole cell pertussis vaccine in laboratory animals. To this end
6 vaccines, both including 'normal' and 'abnormal' products with respect to
levels of Pertussis toxin and/or potency were tested. The study included
the Mouse Weight Gain (MWG) test ; the current WHO-recommended bioassay to
evaluate overall pertussis toxicity and four specific test systems ; the
Leukocytosis Promoting Factor (LPF) test, the Histamine Sensitizing Factor
(HSF) test, and the Chinese Hamster Ovary (CHO) clustering test to estimate
pertussis toxine (PT) levels and the Limulus Amoebocyte Lysate (LAL) test to
evaluate endotoxin levels. In addition participants were also asked to
estimate potency by the Mouse Protection test according to Kendrick (MP).
Fourteen laboratories in various countries participated in the study. In
almost all participating laboratories MWG test was not very accurate in
evaluating the overall toxicity of whole cell pertussis vaccines. In
addition, statistical significant interlaboratory variation was frequently
seen. The specific toxicity tests (LPF, HSF, CHO and LAL test) appeared to
be more accurate, but large interlaboratory variation was seen,
statistically significant at p<0.05 for LPF test, CHO test and LAL test.
Significant variation in test results also occurred in the potency test. In
addition, the discriminative power of the MP test between potent and
non-potent products was low. It is concluded that, on the condition of
optimization and stringent standardization, HSF and CHO test and in
particular LPF test might be more appropriate to assess PT activity than the
MWG test. An inhibition ELISA has been used to estimate levels of PT. This
test could be of value for prescreening purposes. The LAL test should be
used to estimate endotoxin activity. The value of the MP test, as a model
to assess potency, is disputed.

Bij veertien laboratoria in dertien landen heeft een
"collaborative study" plaatsgevonden om de relevantie van 5 testsystemen
voor de controle op de toxiciteit van whole-cell kinkhoest vaccin in muizen
te onderzoeken. Daarvoor werden 6 produkten, die verschillen in toxiciteit
en werkzaamheid, getest. De studie omvat de volgende testsystemen: de Mouse
Weight Gain (MWG) proef, dat is de huidige officieel verplichte proef ter
controle van toxiciteit van kinkhoest en vier specifieke testsystemen ; de
Leukocytose Promoting Factor (LPF) test, de Histamine Sensitizing Factor
(HSF) test en de Chinese Hamster Ovary (CHO) test om pertussis toxine (PT)
waarden te bepalen en de Limulus Amoebocyte Lysate (LAL) test om endotoxine
waarden te meten. Tevens werd aan de deelnemers gevraagd om de werkzaamheid
van de produkten te bepalen door de Mouse Potency (MP) test, volgens
Kendrick. De MWG test bleek in bijna alle deelnemende laboratoria niet
gevoelig genoeg om de totale toxiciteit van whole-cell pertussis te meten.
Bovendien werden er regelmatig tussen de laboratoria statistisch
significante verschillen aangetoond. De specifieke toxiciteitstesten (LPF,
HSF, CHO en LAL test) bleken duidelijk nauwkeuriger, maar tussen de
laboratoria werd voor de LPF test, CHO test en LAL test grote variatie
aangetoond, statistisch significant bij p<0.05. Significante variatie in
onderzoeks- resultaten kwamen eveneens bij MP test voor. Bovendien was het
vermogen van de MP test om onderscheid te kunnen maken tussen produkten met
hoge en lage werkzaamheid klein. De conclusie is, dat onder de voorwaarde
van verfijning en vergaande standaardisatie, de HSF en CHO test en in het
bijzonder de LPF test een meer geschikte test is om biologisch actief PT te
bepalen dan de MWG test. Om het PT gehalte te bepalen werd een inhibitie
ELISA gebruikt Deze test kan voor pre-screening doeleinden van belang zijn.
De LAL test zou gebruikt moeten worden om de endotoxine activiteit te meten.
De betekenis van de MP test, als model om de werkzaamheid te bepalen, wordt
ter discussie gesteld.
